نخستین ارزیابی تنوع ژنتیکی ایزوله‌های انسانی ژیاردیا دئودنالیس با استفاده از تکنیک Multilocus Sequence Typing (MLST) در استان خوزستان

چکیده

مقدمه: ژیاردیا دئودنالیس یکی از مهمترین عوامل انگلی منتقله از مصرف آب و مواد غذایی آلوده می‌باشد. از عوامل مؤثر در بیماریزایی این انگل، ویژگیهای ژنتیکی آن می‌باشد. بررسی حاضر اولین مطالعه تعیین ژنوتایپ ژیاردیا دئودنالیس انسانی با استفاده از تکنیک تایپینگ چندین جایگاه (MLST) از ژنهای Housekeeping است که در استان خوزستان انجام شده است.

مواد و روش‌‌‌ها: DNA ژنومی‌ کیست‌های تخلیص شده از 129 نمونه مدفوع انسانی استخراج و با تکثیر قطعه bp350 از ژن SSU-rRNA ژیاردیا دئودنالیس تأیید گردید. Nested-PCR با هدف ژنهای tpi و β-giardin و Semi-Nested PCR ژن gdh به ترتیب جهت تکثیر قطعات bp530، bp511 و bp432 از ژنهای فوق انجام و قطعات تکثیر یافته تعیین توالی شدند. سپس اسمبلیج‌ها و ساب اسمبلیج‌ها با تجزیه و تحلیل فیلوژنی شناسایی شدند.

نتايج: از مجموع 29 نمونه‌‌ای که در یکی از سه ژن تعیین توالی شدند، 23 نمونه مربوط به اسمبلیج A (31/79 درصد) و 6 نمونه متعلق به اسمبلیجB (69/20 درصد) بودند. در این مطالعه بین اسمبلیج‌ها با علایم بالینی و محل سکونت ارتباط معنی‌داری وجود نداشت. تجزیه و تحلیل فیلوژنی، ساب اسمبلیج‌های AII و BIV را نشان داد.

نتیجه‌گیری: نتایج اسمبلیج A و ساب اسمبلیج AII را بهعنوان ژنوتایپ غالب استان خوزستان نشان داد و شناسایی ساب اسمبلیج‌های AII و BIV، انتقال از مسیر آنتروپونوتیک را پیشنهاد نمود. تعیین نقش حیوانات در پویایی انتقال ژیاردیازیس نیازمند مطالعات بیشتر و دقیق‌تر می‌باشد.
