بررسی اثر هیدروژل کربوکسی متیل سلولز-ژلاتین حاوی عصاره گیاه پنیرک بر ترمیم زخم پوستی: مطالعه پیش بالینی

مقدمه: ترمیم زخمهای پوستی چالش بالینی مهمی است و پانسمانهای زیستسازگار حاوی ترکیبات طبیعی، بهبود را تسریع میکنند. پنیرک (Malva sylvestris) با خواص ضدالتهابی، آنتیاکسیدانی و ضدمیکروبی، گزینهای مناسب برای پانسمان زیستی است. هدف، سنتز و ارزیابی هیدروژل کربوکسیمتیل سلولز-ژلاتین حاوی عصاره هیدروالکلی پنیرک (CMC‑Gel‑Malva) و بررسی اثر آن بر ترمیم زخم در مدل حیوانی میباشد.
مواد و روشها: هیدروژل CMC‑Gel‑Malva سنتز شد. ویژگیهای فیزیکوشیمیایی (مورفولوژی با SEM، جذب آب، زیستتخریبپذیری و pH) ارزیابی گردید. زیستسازگاری خونی با همولیز و شاخص لختهشدن (BCI) و زیستسازگاری سلولی با MTT روی فیبروبلاست 3T3 انجام شد. اثر ترمیمی در مدل زخم موش صحرایی در گروههای کنترل منفی، CMC‑Gel و CMC‑Gel‑Malva طی ۱۴ روز بررسی و درصد بستهشدن زخم و شاخصهای هیستوپاتولوژیک (H&E) ارزیابی شد.
نتایج: همولیز هر دو هیدروژل زیر ۵٪ (بهترتیب ۴٫۱۵٪ و ۴٫۴۰٪) بود BCI در CMC‑Gel (۴۶٫۲۰٪) کمتر از CMC‑Gel‑Malva (۳۱٫۱۰٪) اما هر دو نسبت به کنترل (۱۰۰٪) کاهش معنیداری داشتند. افزودن پنیرک جذب آب را افزایش (۶۵٫۵۳ در برابر ۵۷٫۲۷ در مرحله آخر) و تخریب اولیه را تسریع کرد. MTT در ۷۲ ساعت، زندهمانی بالاتر برای CMC‑Gel‑Malva (۱۰۷٫۲٪ در برابر ۹۹٫۲٪) نشان داد. بستهشدن زخم در روز ۱۴ در گروه CMC‑Gel‑Malva (۸۵٫۴۴٪) بهطور معنیداری بیشتر از CMC‑Gel (۷۸٫۲۲٪) بود. هیستوپاتولوژی گروه CMC‑Gel‑Malva حاکی از کاهش التهاب، بازسازی فولیکولها و غدد چربی، افزایش عروق و کلاژنزایی منظم بود.
نتیجهگیری: هیدروژل CMC‑Gel‑Malva دارای زیستسازگاری عالی، جذب آب بالا و اثر ترمیمکنندگی مطلوب است و میتواند بهعنوان پانسمان زیستی مؤثر برای زخمهای مزمن و سوختگی درجه دو پیشنهاد شود.
