اثر بدون بار کردن مکانیکی اندام تحتانی بر بیان ژن‌های MST1 و آتروژین1 عضلات نعلی و کف‌پایی موش‌های صحرایی ویستار

چکیده

مقدمه: کیناز شبه Ste20 پستانداران  (The mammalian Ste20-like kinase) با فعال کردن لیگاز مسیر یوبی کوئیتینی آتروژین1 در آتروفی عصبی درگیر میباشد. از این رو، هدف از پژوهش حاضر بررسی بیان mRNA ژنهای MST1 و آتروژین1 و مقایسه آنها در عضلات نعلی و کف پایی در موشهای صحرایی قرار گرفته شده در حالت تعلیق اندام تحتانی بود.

مواد و روش‌ها: در این پژوهش 10 سر موش صحرایی نژاد ویستار با میانگین سنی 6-4 ماه به طور تصادفی در دو گروه تعلیق اندام تحتانی و کنترل قرار گرفتند (5 سر موش صحرایی در هر گروه). نمونههای گروه تعلیق به مدت 14 روز به روش آویزان کردن از دم نگهداری شدند و نمونههای گروه کنترل در این مدت زندگی عادی را سپری میکردند. پس از 14 روز موشها تشریح شده و عضلات نعلی و کف پایی آنها جمعآوری شد و پس از وزن کردن عضله در نیتروژن مایع نگهداری شدند. سپس میزان بیان mRNA ژنهای MST1 و آتروژین1 در دو عضله اندازهگیری شد.

نتایج: نتایج پژوهش حاضر نشان داد که پس از 14 روز تعلیق اندام تحتانی وزن نسبی هر دو عضله نعلی و کف پایی کاهش یافته بود ولی این کاهش در کف پایی معنیدار نبود. علاوه بر این بیان mRNA  ژن MST1 (021/0=P) افزایش معنیدار و آتروژین1 (056/0=P) در عضله نعلی افزایش یافت. همچنین، هر دو MST1 (071/0=P) و آتروژین1 (33/0=P) افزایش غیر معنیداری را در عضله کف پایی نشان دادند. مقایسه بیان mRNA  ژنها در دو عضله نشان داد که بیان mRNA MST1 در نعلی بیشتر از عضله کف پایی بود ولی افزایش بیشتری در mRNA آتروژین1 در عضله کف پایی نسبت به نعلی نشان داده شد.

نتیجه‌گیری: با توجه به نتایج پژوهش حاضر میتوان اظهار داشت که MST1 میتواند یکی از اهداف اصلی کنترل آتروفی در شرایط بدون بار شدن مکانیکی همانند بستری شدن و سفرهای فضایی در نظر گرفته شود. هر چند که اهداف پایین دست MST1 در دو عضله میتواند متفاوت باشد.
